Overview:

Molecular Biology Grade, recombinant subtilisin-related serine protease K (Proteinase K) from Parengyodontium album (Tritirachium album) is expressed in Pichia pastoris and purified by extensive chromatography. This broad-spectrum endopeptidase is widely used for digestions of proteins, including DNase and RNase, in nucleic acid preparations.


Storage and Stability:

Dry powder status -20°C valid for 24 months; the enzyme powder should be dissolved into the appropriate volume, 2-8°C short-term storage, -20°C long-term storage. Rehydrate the enzyme powder using nuclease-free water, and it is recommended to store the Proteinase K solution at 2-8°C.


Activity:

One unit of Proteinase K hydrolyzes urea-denatured hemoglobin producing colour equivalent of 1μmol tyrosine per 1 min at 37oC and pH 7.5 (Folin & Ciocalteu’s method), 1U = 1mAnsonU

No detectable endonuclease, exonuclease, DNase or RNase contaminating activities


Buffers:

For immediate use, Protease K may be dissolved in a solution of 50 mM Tris-HCl, pH 7.8, 3 mM CaCl2.
For long-term storage, Protease K may be stored in a solution of 50 mM Tris-HCl, pH 7.8, 3 mM CaCl2, 50% glycerol at -20°C.


Purity:

The purity of Proteinase K was determined to be >90% by densitometry
